Germinal centers are the engines of affinity maturation. A greater understanding of how to track, characterize, and induce Tfh cell responses could greatly contribute to the realization of new vaccines. Following up on our recent work on signals and circuits involved in Tfh differentiation, we have recently spent extensive effort studying: (1) How can germinal center responses to a vaccine be maximized? (2) What signals can induce human Tfh differentiation? (3) How can antigen-specific GC Tfh cells be tracked and quantified? (4) Can germinal center responses be tracked longitudinally in an animal model? (5) What can we learn about GC Tfh cells in human disease states that can teach us about optimal T cell help to B cells?
